VSV-IL-15 is an oncolytic virus therapy based on vesicular stomatitis virus (VSV) engineered to express a highly secreted form of human interleukin-15 (IL-15). This recombinant virus is designed to combine the direct lytic activity of VSV against tumor cells with the immunostimulatory properties of IL-15, which enhances natural killer (NK) and CD8+ T cell responses. Preclinical models have demonstrated that VSV-IL-15 can improve anti-tumor immunity, increase survival compared to VSV alone, and is associated with increased infiltration and activation of immune effector cells in the tumor microenvironment[1][4]. In combination regimens involving NKT cell immunotherapy and PD-1 blockade, VSV-IL-15 led to durable tumor regression and immune memory response in mouse models[4].
