Overview

VSV-M51-NIS is an engineered oncolytic vesicular stomatitis virus (VSV) designed for cancer therapy. It features a specific methionine deletion at residue 51 (M51) of the matrix (M) protein, which prevents the virus from blocking the host cell's nuclear-to-cytoplasmic transport of interferon-beta (IFN-β) mRNA. This modification attenuates the virus in normal cells with intact interferon pathways while allowing selective replication and oncolysis in malignant cells with defective antiviral signaling. Additionally, the virus is engineered to express the thyroidal sodium iodide symporter (NIS), which enables non-invasive imaging of viral distribution using radioisotopes and potentially facilitates targeted radiovirotherapy. It has shown preclinical efficacy in models of endometrial cancer and multiple myeloma.
