VSV-mIFNβ-NIS is a genetically engineered recombinant vesicular stomatitis virus (VSV) that encodes murine interferon-β (mIFNβ) and the sodium iodide symporter (NIS). Designed as an oncolytic virotherapy, it selectively destroys tumor cells with defective interferon pathways while sparing normal tissue. The expression of mIFNβ enhances tumor selectivity and antitumor immune responses and limits VSV replication in healthy cells, increasing safety. The NIS reporter gene enables noninvasive imaging of viral infection and intratumoral distribution. Preclinical studies have demonstrated robust tumor debulking and immune-mediated tumor clearance in murine models, especially in multiple myeloma, with intravenous administration leading to rapid oncolytic activity and sustained antitumor immunity. Clinical studies have been initiated for related constructs in multiple myeloma, lymphoma, acute myeloid leukemia, and endometrial cancer.
