VT30

VT30 is a topical prodrug of VT10, which acts as an inhibitor of the class I phosphatidylinositol-4,5-bisphosphate 3-kinase catalytic subunit alpha (PIK3CA). It is designed for local delivery to cutaneous lesions and aims to minimize systemic side effects by being metabolized in the skin to its active form. The drug was developed primarily for the treatment of cutaneous venous malformations, lymphatic malformations, or mixed venolymphatic malformations associated with PIK3CA or TEK gene mutations. Clinical studies demonstrated that high concentrations of the active drug are achieved locally in treated tissue with no detectable systemic exposure. The most common adverse event reported was a mild-to-moderate local rash at higher concentrations. Development has been discontinued after reaching Phase 2 trials[1][2][4][7].
