VTP-600 is a novel prime-boost therapeutic cancer vaccine under clinical investigation as an immunotherapy for cancer, initially developed for non-small cell lung cancer (NSCLC). It leverages a proprietary heterologous viral vector platform combining a chimpanzee adenovirus (ChAdOx1) and Modified Vaccinia Ankara (MVA), each engineered to express the tumor-associated antigens MAGE-A3 and NY-ESO-1. By delivering these antigens to antigen presenting cells, VTP-600 is designed to stimulate the immune system to generate cytotoxic CD8+ T cells capable of targeting and killing cancer cells expressing these antigens. The vaccine regimen involves an initial 'prime' dose delivered with the ChAdOx1 vector, followed by a booster with MVA-based vectors, tailored based on tumor antigen expression. The therapy is administered intramuscularly and is being evaluated in combination with standard first-line treatments, including chemotherapy and checkpoint inhibitors, in ongoing Phase 1/2a clinical trials in patients newly diagnosed with NSCLC. The approach has the potential to be adapted for other cancers expressing MAGE-A3 and NY-ESO-1 antigens, such as breast, bowel, bladder, and melanoma[1][2][3][5][6][7][8][10].
