VTRU200 is a first-in-class trispecific T-cell engager (TCE) developed by Vitruviae that simultaneously targets high mannose (Man9) oligosaccharides, phosphatidylserine (PS), and CD3. Unlike traditional protein-targeted immunotherapies, VTRU200 targets non-protein markers that are aberrantly expressed on the surface of malignant cells but absent or sequestered in healthy tissues. Man9 glycans arise from errors in glycan assembly in cancer cells, while PS flips to the outer leaflet of the cell membrane during malignant transformation. By dual-targeting these pan-cancer molecular patterns, VTRU200 aims to reduce on-target, off-tumor toxicity and mitigate antigen-negative relapse. Preclinical studies have demonstrated potent anti-tumor activity in acute myeloid leukemia (AML) and various solid tumors, including pancreatic cancer and diffuse intrinsic pontine glioma (DIPG).
