VTS208 is a first-in-class trispecific T-cell engager (TCE) designed to target Claudin 18.2 (CLDN18.2) and Cadherin 17 (CDH17) while simultaneously engaging CD3 on T cells. Developed for the treatment of gastrointestinal (GI) cancers, including gastric cancer (GC), pancreatic ductal adenocarcinoma (PDAC), and colorectal cancer (CRC), VTS208 utilizes a proprietary CD3-binding arm and a bi-paratopic CDH17-targeting design to enhance binding avidity and efficacy against tumors with low antigen expression. By targeting two highly complementary tumor-associated antigens, the drug aims to address tumor heterogeneity and overcome antigen-loss resistance common in single-target therapies. Preclinical data demonstrate robust anti-tumor activity and a favorable safety profile, with an IND application planned for the second half of 2026.
