VTX-0811 is a first-in-class investigational monoclonal antibody developed by Verseau Therapeutics for the treatment of solid tumors. It targets P-selectin glycoprotein ligand 1 (PSGL-1), an adhesion molecule involved in immune cell trafficking. By binding to PSGL-1, VTX-0811 repolarizes immunosuppressive tumor-associated macrophages into pro-inflammatory, anti-tumorigenic macrophages without affecting selectin and VISTA-mediated functions of PSGL-1. This transformation initiates a coordinated immune response that attracts and activates cancer-fighting immune cells within the tumor microenvironment, offering a potentially broader and more potent antitumor effect compared to current T cell-based immunotherapies[1][2][5]. Preclinical studies have shown efficacy in human cells and animal models, with favorable safety profiles leading to FDA clearance for clinical trials[2][4].
