Overview

VTX2735 is a novel, highly potent and selective oral small molecule inhibitor of NLRP3 (NACHT, LRR and PYD domains-containing protein 3), developed for the treatment of systemic inflammatory diseases. Its mechanism of action involves inhibition of the NLRP3 inflammasome pathway, which plays a key role in mediating inflammation through downstream cytokines such as IL-1β and IL-6. Clinical proof-of-concept has been established in Phase 2 trials for cryopyrin-associated periodic syndromes (CAPS), including familial cold autoinflammatory syndrome (FCAS), where it demonstrated significant improvements in disease activity and reduction in inflammatory biomarkers. The drug is also being developed for recurrent pericarditis and other cardiovascular or metabolic conditions characterized by aberrant NLRP3 activation. Developed by Ventyx Biosciences (originator Zomagen Biosciences), it is administered orally as a tablet or suspension[1][4][7].
