Drug intelligence / Profile preview
VU0134992 is a selective, orally active small-molecule pore blocker of the inwardly rectifying potassium channel Kir4.1 (encoded by KCNJ10) and heteromeric Kir4.1/5.1 channels. Discovered through high-throughput screening of the Vanderbilt Institute of Chemical Biology library, VU0134992 exhibits selectivity for homomeric Kir4.1 channels over other Kir channel subtypes, including Kir1.1, Kir2.1, and Kir2.2. In preclinical animal models, oral administration of VU0134992 induces dose-dependent diuresis, natriuresis, and kaliuresis, and has been shown to acutely reduce serum potassium levels in hyperkalemic rats with reduced kidney function. It serves as a valuable pharmacological tool compound to investigate the physiological roles of Kir4.1 in renal function, astrocyte physiology, and its potential as a therapeutic target for hypertension and hyperkalemia.
