Vudalimab is a bispecific monoclonal antibody engineered to simultaneously target the immune checkpoint receptors programmed cell death protein 1 (PD‑1) and cytotoxic T‑lymphocyte-associated antigen 4 (CTLA‑4). By blocking both PD‑1 and CTLA‑4, vudalimab is designed to promote tumor-selective T-cell activation, enhancing anti-tumor immune responses. The drug uses Fc engineering technology (XmAb platform) to improve selectivity for T cells with high expression of both checkpoints, potentially improving the therapeutic index compared to combination immunotherapies. Vudalimab has been investigated primarily for solid tumors including metastatic castration-resistant prostate cancer, non-small cell lung cancer, biliary cancer, gynecological cancers, and other advanced malignancies. It was developed by Xencor[1][3][5][6].
