Overview

VUF11207 is a small molecule agonist of the atypical chemokine receptor 3 (ACKR3), formerly known as CXCR7. It is currently being investigated as an immunomodulatory agent for the treatment of glioblastoma (GBM). VUF11207 works by activating ACKR3 to inhibit the release of the chemokine CXCL12 from tumor cells, which prevents the recruitment and activation of glioblastoma-associated macrophages (GAMs). This inhibition leads to the downregulation of PD-L1 on GAMs and promotes the infiltration of cytotoxic CD8+ T-cells into the tumor microenvironment. To overcome the blood-brain barrier (BBB), researchers have developed a lipid-based nanoparticle (LNP) formulation of VUF11207 (VUF@LNP) designed for non-invasive intranasal delivery. Preclinical studies have demonstrated that this intranasal delivery method significantly improves survival outcomes in GBM models, particularly when used in combination with PD-L1 blockade therapy.
