Overview

VV169 is an experimental in vivo CAR T-cell therapy being developed by Vyriad for the treatment of relapsed or refractory multiple myeloma. It consists of a single intravenous administration of a lentiviral vector (LV-169) engineered with advanced G protein retargeting to selectively transduce T cells in vivo with a chimeric antigen receptor (CAR) that recognizes B-cell maturation antigen (BCMA) on malignant plasma cells, thereby converting the patient’s own T cells into BCMA-directed CAR T cells inside the body and eliminating the need for ex vivo cell manufacturing.[1][3][6][7][8] Preclinical studies in humanized mouse models of disseminated myeloma showed that a single dose achieved complete clearance of tumors in 100% of treated animals at all tested dose levels, with durable responses, resistance to tumor re-challenge, and an apparently favorable cytokine profile with minimal inflammatory cytokine elevation.[1][3][6][7] Vyriad plans to initiate first-in-human clinical trials of VV169 in the U.S. in patients with multiple myeloma, leveraging this platform as one of the earliest clinical implementations of an off-the-shelf in vivo CAR T approach.[1][3][6][7][8]
