VVD-159642 + trametinib is an investigational combination therapy being evaluated for the treatment of advanced solid tumors, especially RAS-driven cancers. VVD-159642 is a novel, oral small molecule inhibitor developed to selectively prevent rat sarcoma viral oncogene (RAS) activation of the phosphatidylinositol 3-kinase alpha (PI3Kα) pathway, thereby blocking oncogenic signaling that is vital in cancer cell growth and survival while sparing normal cellular function. Trametinib is an approved small molecule inhibitor of the mitogen-activated protein kinase kinase (MEK1/2) that acts within the MAPK pathway. The combination is tested to explore synergistic inhibition of RAS/MAPK and RAS/PI3K pathways, aiming to overcome resistance mechanisms and improve efficacy in patients with advanced malignancies. Currently, the combination is in phase I clinical trials, administered orally in 21-day cycles, with preliminary endpoints focusing on safety, tolerability, pharmacokinetics, and anti-tumor activity[1][2][3][4][6][7][8][9].
