Overview

VVD-159642 + sotorasib is an oral combination therapy under investigation for the treatment of advanced solid tumors driven by RAS mutations. VVD-159642 is a small molecule inhibitor that selectively blocks activation of the PI3K-alpha pathway by RAS proteins, aiming to inhibit oncogenic signaling crucial to solid tumor growth without affecting normal cellular function. Sotorasib is a small molecule targeting KRAS G12C, covalently inhibiting mutant KRAS, and is approved as a targeted therapy for specific cancers. This combination is being tested to determine safety, pharmacodynamics, pharmacokinetics, and preliminary anti-tumor activity, based on preclinical rationale that dual targeting of RAS-driven and MAPK pathways may increase efficacy or overcome resistance seen with single agents. Both agents are administered orally.
