Overview

vvDD-CCL5 is an oncolytic vaccinia virus (VACV) engineered for enhanced tumor selectivity and immunogenicity. The "vvDD" backbone refers to a "double-deleted" vaccinia virus where the genes for thymidine kinase (TK) and vaccinia growth factor (VGF) have been removed to restrict viral replication to rapidly dividing malignant cells. This specific construct is further "armed" with a transgene encoding the chemokine CCL5 (also known as RANTES). CCL5 expression within the tumor microenvironment is intended to recruit immune effector cells, such as T cells and dendritic cells, thereby converting "cold" tumors into "hot" tumors and enhancing the anti-tumor immune response. Research has demonstrated its efficacy in murine models of colon cancer and malignant mesothelioma, particularly when combined with other immunotherapies like IL-2 or checkpoint inhibitors.
