vvDD-IL-2 is an oncolytic virus therapy candidate consisting of a genetically modified, replication-restricted vaccinia virus (Western Reserve strain) engineered to express human interleukin-2 (IL-2). It is designed to selectively infect and lyse tumor cells (oncolysis) while simultaneously delivering IL-2 directly into the tumor microenvironment. The expressed IL-2, often in a GPI-anchored form to limit systemic exposure, stimulates the activation and proliferation of cytotoxic T-lymphocytes and natural killer (NK) cells. This dual approach aims to convert immunologically 'cold' tumors into 'hot' tumors, enhancing the anti-tumor immune response. It is being developed through a collaboration between Charité-Universitätsmedizin Berlin and the University of Pittsburgh for the treatment of solid tumors, specifically pancreatic ductal adenocarcinoma (PDAC).
