vvDD-scIL-12-R-IL-2-RG is an engineered oncolytic vaccinia virus (oVV) designed to deliver a membrane-tethered fusion cytokine consisting of single-chain interleukin-12 (scIL-12) and interleukin-2 (IL-2). The virus utilizes the vvDD backbone, which is double-deleted for thymidine kinase (TK) and vaccinia growth factor (VGF) to enhance tumor-selective replication. By expressing these cytokines as a tethered fusion protein on the surface of infected tumor cells, the agent aims to stimulate a potent localized immune response—characterized by increased Th1 chemokines, IFN-γ production, and CD8+ T cell infiltration—while minimizing the systemic toxicities associated with recombinant cytokine therapy. Preclinical studies in murine models of Lewis lung carcinoma, renal adenocarcinoma (Renca), and colon adenocarcinoma (MC38) have demonstrated robust antitumor activity, with high rates of complete regression and the induction of durable, tumor-specific immunity.
