VVN432 is a small molecule, non-steroidal Janus kinase (JAK) inhibitor developed by VivaVision Biotech for the treatment of chronic rhinosinusitis (CRS), including patients with or without nasal polyps. Formulated as a nasal spray, VVN432 is designed to provide localized anti-inflammatory effects by inhibiting the JAK signaling pathway, which is a central mediator for various pro-inflammatory cytokines involved in the pathogenesis of inflammatory sinus diseases. By utilizing a topical intranasal delivery system, the therapy aims to achieve high local concentrations in the sinonasal mucosa while minimizing the systemic exposure and associated side effects typically seen with oral JAK inhibitors. The drug is currently undergoing Phase 1 clinical evaluation to assess its safety, tolerability, and pharmacokinetics in both healthy volunteers and CRS patients.
