Overview

VVY248 is a small molecule inhibitor of glutaminyl-peptide cyclotransferase (QPCT) and glutaminyl-peptide cyclotransferase-like (QPCTL) enzymes. Developed by Vivoryon Therapeutics (formerly Probiodrug), it is currently in the Pre-IND stage for the treatment of kidney disorders characterized by fibrosis and inflammation. The drug targets the enzymatic activity of QPCT and QPCTL, which are involved in the post-translational modification of proteins, specifically the formation of pyroglutamate (pGlu) residues. This modification is linked to the stability and aggregation of proteins involved in inflammatory and fibrotic pathways. VVY248 represents a follow-on program to Vivoryon's lead candidate, varoglutamstat, focusing specifically on renal indications where fibrosis and inflammation are primary drivers of disease progression.
