Overview
VX-128 is a small molecule, highly potent, and selective inhibitor of the voltage-gated sodium channel Nav1.8 (SCN10A). Developed by Vertex Pharmaceuticals, it was intended for the treatment of acute pain. Nav1.8 is primarily expressed in peripheral sensory neurons and plays a critical role in pain signaling. VX-128 was designed to offer an improved profile over earlier candidates like VX-150. However, its clinical development was terminated during Phase 1 dose-escalation studies in healthy volunteers due to safety concerns, specifically the occurrence of skin rashes in approximately 19% of participants and a case of angioedema.
