VX-222 + telaprevir is an investigational oral combination of two direct-acting antivirals developed for the treatment of genotype 1 chronic hepatitis C virus (HCV) infection. **VX-222** is a small molecule non-nucleoside inhibitor that selectively targets the HCV NS5B polymerase, a key enzyme necessary for viral RNA replication. **Telaprevir** is a small molecule inhibitor of the HCV NS3/4A protease, another essential viral enzyme that cleaves the viral polyprotein, enabling viral maturation and replication. Administered together, these agents act on distinct steps of the viral lifecycle to suppress HCV replication more effectively. The combination has been tested in various multi-drug regimens (including with pegylated interferon and ribavirin) in phase 2 trials.
