Overview

VX-222 is a selective, non-nucleoside inhibitor of HCV NS5B polymerase, while telaprevir is a specific, reversible, covalent inhibitor of the HCV NS3/4A protease. Ribavirin is a nucleoside analog with broad-spectrum antiviral activity, commonly used as part of HCV treatment. The combination aims to target two essential viral enzymes, increasing the barrier to resistance and treatment efficacy. This regimen has been evaluated in phase 2 studies (notably the ZENITH trial), showing substantial antiviral activity and sustained virologic response, with improved tolerability when peginterferon is excluded[1][2][4][7].
