VX-828 + tezacaftor + deutivacaftor is an investigational triple combination therapy for cystic fibrosis. VX-828 is a next-generation CFTR corrector, tezacaftor is an established CFTR corrector, and deutivacaftor (VX-118) is a next-generation CFTR potentiator. This regimen targets the underlying defect in CFTR protein caused by CF mutations—primarily increasing CFTR protein quantity and functional activity at the epithelial surface, aiming to achieve near-normal chloride channel function. The combination is designed to treat patients carrying F508del and minimal function CFTR mutations not responsive to approved triple modulators. The primary indication is cystic fibrosis, specifically in adults with F508del/MF genotypes, and it's being evaluated for improved efficacy over existing standards like elexacaftor/tezacaftor/ivacaftor and vanzacaftor/tezacaftor/deutivacaftor regimens[4][1][3].
