VX-883 is a novel, small-molecule antibiotic developed by Vertex Pharmaceuticals. It was designed as a dual-mechanism inhibitor targeting both DNA gyrase (specifically the GyrB subunit) and topoisomerase IV (specifically the ParE subunit). This dual-targeting approach was intended to provide broad-spectrum activity against Gram-positive and Gram-negative bacteria, including multi-drug resistant strains such as methicillin-resistant *Staphylococcus aureus* (MRSA) and vancomycin-resistant enterococci (VRE). By binding to the ATP-binding sites of these essential enzymes, VX-883 inhibits bacterial DNA replication and transcription. Although Vertex planned to initiate clinical trials in 2007, the compound did not progress into late-stage development and was subsequently discontinued from the company's pipeline.
