**Vx1000R** is a mouse anti-human CD47 monoclonal antibody developed by Vasculox Inc. (now part of Arch Oncology). It targets the CD47 protein, which binds to signal-regulatory protein alpha (SIRPα), a receptor on the surface of macrophages. By blocking this interaction, Vx1000R induces macrophage-mediated phagocytosis of tumor cells, particularly effective in killing multiple myeloma (MM) cells when macrophages are present. In 3D tissue-engineered bone marrow models, it has shown a 75% killing rate of MM cells compared to controls. Vx1000R primarily acts by promoting macrophage-dependent clearance of cancer cells, making it a promising therapeutic agent in the treatment of hematological malignancies like multiple myeloma.
