VXX-301 is a preclinical, synthetic peptide-based active immunotherapy (vaccine) targeting pathologic forms of microtubule-associated protein tau for the treatment and prevention of Alzheimer’s disease and other tauopathies. It comprises a mixture of two tau-targeting peptides (p5555kb and p5187kb), each conjugated to Vaxxinity’s proprietary T-helper carrier peptide, and is formulated with CpG and AdjuPhos adjuvants to enhance immunogenicity.[1][6][9] The multi-epitope design targets N‑terminal and repeat domains of tau, inducing predominantly IgG1 antibodies that bind multiple tau isoforms and conformations, block tau aggregation and cellular uptake in vitro, and improve tau-dependent phenotypes and survival in tau transgenic mouse models, supporting its development as a disease-modifying tau vaccine for Alzheimer’s disease and other tau-driven neurodegenerative disorders.[1][6][7]
