VXX-401 is a synthetic peptide-based vaccine developed to lower low-density lipoprotein cholesterol (LDL-C) by inducing the body to produce antibodies against proprotein convertase subtilisin/kexin type 9 (PCSK9). By targeting PCSK9, these antibodies prevent PCSK9 from inhibiting the LDL receptor, thereby enhancing LDL clearance from the bloodstream. Preclinical studies in nonhuman primates have shown that VXX-401 induces robust and durable anti-PCSK9 antibody responses, resulting in sustained reductions of LDL-C by approximately 30–50%. The vaccine has demonstrated a favorable safety profile with only mild injection site reactions observed. VXX-401 is being developed as an alternative to statins and monoclonal antibody therapies for hypercholesterolemia and aims to provide a more accessible and convenient option for preventing atherosclerotic cardiovascular disease (ASCVD). Vaxxinity is the developer and manufacturer. VXX-401 is currently in Phase 1 clinical trials for safety and tolerability in humans.
