Drug intelligence / Profile preview
Vyxeos (CPX-351) is a liposomal co-formulation of the antimetabolite cytarabine and the anthracycline daunorubicin in a fixed 5:1 molar ratio. This specific ratio has been shown to be synergistic in vitro. The liposomal delivery system (CombiPlex platform) is designed to maintain this ratio in vivo and preferentially deliver the drugs to the bone marrow, where they are taken up by leukemia cells. Cytarabine inhibits DNA synthesis by competing with cytidine for incorporation into DNA, while daunorubicin inhibits DNA topoisomerase II, leading to DNA strand breaks. It is FDA-approved for newly diagnosed therapy-related AML (t-AML) or AML with myelodysplasia-related changes (AML-MRC). Fred Hutchinson Cancer Center is investigating its use in a Phase II trial for patients with high-grade myeloid neoplasms (including MDS-EB2) with measurable residual disease (MRD) prior to allogeneic hematopoietic cell transplantation.
