Drug intelligence / Profile preview
Vyxeos (CPX-351) is a liposomal formulation of the chemotherapy agents cytarabine and daunorubicin, encapsulated in a fixed 5:1 molar ratio. This specific ratio is designed to maximize synergistic antineoplastic activity while minimizing toxicity. Daunorubicin, an anthracycline, acts by intercalating into DNA and inhibiting DNA topoisomerase II, while cytarabine, a nucleoside analog, inhibits DNA synthesis by competing with cytidine for incorporation into DNA. Originally developed by Celator Pharmaceuticals and later acquired by Jazz Pharmaceuticals, Vyxeos is FDA-approved for the treatment of adults with newly diagnosed therapy-related acute myeloid leukemia (t-AML) or AML with myelodysplasia-related changes (AML-MRC). The drug is also being investigated in pediatric and young adult populations for a variety of relapsed or refractory hematologic malignancies, including acute lymphoblastic leukemia and various types of lymphoma, through clinical trials such as those led by Cincinnati Children's Hospital Medical Center.
