W-CCL27 CAR-T is an experimental chimeric antigen receptor (CAR) T-cell therapy designed to target CCR10, a chemokine receptor highly expressed on multiple myeloma (MM) tumor cells. Developed by researchers at the University of California, San Francisco (UCSF), this therapy utilizes a structure-guided mutant of the natural ligand CCL27 as the binding domain. Specifically, the addition of a single tryptophan (W) residue to the N-terminus of CCL27 significantly enhances binding avidity and cytotoxic efficacy compared to wild-type CCL27. Preclinical studies presented at ASH 2024 demonstrate that W-CCL27 CAR-T cells exhibit potent anti-tumor activity in multiple myeloma models, comparable to BCMA-targeted CAR-T therapies, while showing minimal toxicity to hematopoietic stem cells. The therapy is being investigated as a potential treatment for patients who relapse after anti-BCMA therapies.
