Overview

W-GTF01 is a novel, small molecule inhibitor that selectively targets MGAT1, a glycosyltransferase responsible for catalyzing the N-acetylglucosamine addition to CD73, facilitating its dimerization and surface translocation on tumor cells. In preclinical models of triple-negative breast cancer (TNBC), W-GTF01 blocks MGAT1-catalyzed CD73 glycosylation. This leads to decreased CD73 function on the tumor cell membrane, mitigating immunosuppressive adenosine production, restoring CD8+ T cell responses, and thus sensitizing refractory tumors to immune checkpoint blockade (specifically anti-PD-L1 therapy)[1][3][4][7][10]. W-GTF01 is described as a potential approach to counteract tumor immune evasion and enable effective cancer immunotherapy, especially in so-called immune-cold tumors[3][7][10].
