W0180 is a first-in-class monoclonal antibody that targets VISTA (V-domain Ig suppressor of T cell activation), a negative checkpoint regulator of T cell response. By binding to the extracellular domain of VISTA on myeloid cells and T cells within the tumor microenvironment, W0180 inhibits VISTA-mediated immunosuppression and has the potential to enhance antitumor immune responses. It is being developed as both a monotherapy and in combination with pembrolizumab (an anti-PD-1 therapy) for patients with advanced or metastatic solid tumors. The drug is currently in Phase I clinical trials. Increased expression of VISTA has been observed following treatment with other checkpoint inhibitors such as anti-PD1/L1 and anti-CTLA4 therapies, suggesting that targeting VISTA may help overcome resistance mechanisms to existing immunotherapies[1][2][3][4][5][7].
