W035 is a novel, functionalized β-lactam-based small molecule developed as a multi-target anti-melanoma agent. Synthesized through diversity-oriented synthesis, it demonstrates potent cytotoxicity against human and murine cutaneous melanoma cell lines. Its mechanism of action involves the simultaneous inhibition of the MAPK, PI3K, and JAK/STAT signaling pathways, leading to the downregulation of key melanoma signaling nodes. Phenotypically, W035 disrupts redox homeostasis by reducing intracellular reactive oxygen species, inhibits cell migration and colony formation, and induces apoptosis through the activation of caspase-3/7. Preclinical ADME profiling suggests favorable drug-likeness, including high skin permeability and good gastrointestinal absorption, supporting its potential for both topical and oral administration.
