W146 is a potent and selective small molecule antagonist of the sphingosine-1-phosphate receptor 1 (S1PR1). It is widely utilized as a pharmacological research tool to investigate S1P1 signaling pathways, particularly those governing lymphocyte egress from lymphoid tissues and the maintenance of vascular endothelial barrier function. W146 exhibits high selectivity for S1PR1 (Ki = 18 nM) with no significant activity at S1P2, S1P3, or S1P5 receptors. In preclinical models, administration of W146 has been shown to induce lymphopenia by blocking lymphocyte exit into the blood and lymph, and to increase pulmonary vascular permeability by enhancing capillary leakage. It is not currently under clinical development for any therapeutic indication and is primarily available through chemical suppliers for laboratory research purposes.
