Waglerin-1 is a 22-amino acid peptide toxin originally isolated from the venom of the Southeast Asian Wagler's pit viper (*Trimeresurus wagleri*). It acts as a potent and selective competitive antagonist of the adult-type muscle nicotinic acetylcholine receptor (nAChR), specifically targeting the epsilon (ε) subunit interface. Due to its high affinity for the adult (α1₂β1δε) versus fetal (α1₂β1δγ) nAChR subtypes, it is widely utilized as a pharmacological tool to differentiate between these receptor forms. Additionally, Waglerin-1 has been shown to modulate GABA(A) receptors in a neuron-specific manner, exhibiting both potentiating and suppressing effects. While not an approved therapeutic, its muscle-relaxing properties have led to the development of synthetic derivatives for use in topical cosmetic applications to reduce the appearance of fine lines and wrinkles.
