Overview

WAY170523 is a potent and selective small-molecule inhibitor of matrix metalloproteinase-13 (MMP-13, also known as collagenase-3), with an IC50 of 17 nM. It was originally developed by Wyeth Research (later acquired by Pfizer) using structure-based design and NMR spectroscopy. WAY170523 exhibits high selectivity for MMP-13, showing over 5800-fold selectivity against MMP-1, 56-fold against MMP-9, and over 500-fold against TNF-alpha converting enzyme (TACE). Due to its ability to prevent extracellular matrix degradation, it has been widely investigated in preclinical research for osteoarthritis, rheumatoid arthritis, cancer metastasis, and cardiac dysfunction.
