WC26 is a substituted phenylpiperazine small molecule that acts as a selective partial agonist of the dopamine D3 receptor. Developed as part of the 'WC series' of dopaminergic ligands, it exhibits approximately 50-fold binding selectivity for the D3 receptor over the D2 receptor subtype. In preclinical models of Parkinson's disease, specifically 6-hydroxydopamine (6-OHDA) unilaterally lesioned rats, WC26 has demonstrated significant efficacy in attenuating L-dopa-induced dyskinesia (LID). Mechanistically, it functions as a partial agonist at both D3 (approximately 69% intrinsic activity) and D2 (approximately 30% intrinsic activity) receptors, as measured by adenylyl cyclase inhibition assays. Its ability to reduce abnormal involuntary movements (AIMs) without completely abolishing the therapeutic effects of L-dopa makes it a candidate for managing motor complications in Parkinson's disease.
