Overview

The WCNDD ROCK2 inhibitor program is a small-molecule drug discovery initiative developed by the Warren Center for Neuroscience Drug Discovery (WCNDD) at Vanderbilt University. The program focuses on the development of potent and selective inhibitors of Rho-associated protein kinase 2 (ROCK2), a serine/threonine kinase that serves as a key effector of the RhoA GTPase. ROCK2 is a critical regulator of the actin cytoskeleton, cell polarity, and migration. In neurodegenerative research, ROCK2 inhibition is being investigated for its potential to protect dopaminergic neurons in Parkinson's Disease (PD) by reducing alpha-synuclein aggregation and mitigating neuroinflammation. In oncology, the program targets the role of ROCK2 in promoting the epithelial-mesenchymal transition (EMT), invasion, and metastasis in cancers such as prostate and ovarian cancer. The program is currently in the IND-enabling phase, with candidates being prepared for clinical evaluation.
