WecMix01 is a CRISPR-optimized probiotic consortium consisting of five bacterial strains, including Bifidobacterium animalis WKB148, designed for precision radioprotection and gut microenvironment reprogramming. The consortium is delivered via a pH-gated supramolecular armor composed of Fe³⁺-tannic acid coordination functionalized with hyaluronic acid for CD44 targeting. This biohybrid platform is engineered to protect the intestinal stem cell niche through HDAC3-Notch1 activation, induce IL-22⁺ macrophages to mediate gut-liver crosstalk, and reset the microbial landscape to increase butyrate production. Mechanistically, it exhibits potent reactive oxygen species (ROS) scavenging capabilities and promotes adipocyte browning via PPARγ-UCP1 activation, making it a candidate for treating radiation-induced injury and obesity-associated radiation syndrome.
