WEE1 small interfering RNA is an RNA interference (RNAi) therapeutic designed to silence the expression of the WEE1 kinase. WEE1 is a key regulator of the G2/M cell cycle checkpoint, preventing cells with DNA damage from entering mitosis. In cancers like diffuse pleural mesothelioma (DPM), WEE1 is often overexpressed. Silencing WEE1, particularly in combination with other cell cycle regulators like PKMYT1, induces synthetic lethality by forcing cells into premature mitosis with unrepaired DNA damage, leading to apoptosis. This specific siRNA is being evaluated by researchers at the National Cancer Institute (NCI) within a nanoparticle-hydrogel delivery platform for localized treatment of anatomically restricted cancers.
