Overview

Wetimicin is a semi-synthetic aminoglycoside antibiotic developed by North China Pharmaceutical Group (NCPC). Derived from sisomicin, its chemical structure is 1-N-ethyl-6'-N-methylsisomicin. Wetimicin functions by binding to the 30S subunit of the bacterial ribosome, specifically the 16S rRNA, which interferes with the initiation of protein synthesis and induces translational errors. This leads to the production of nonfunctional or toxic proteins and subsequent bacterial cell death. It exhibits broad-spectrum activity against Gram-negative bacilli, including *Escherichia coli*, *Klebsiella pneumoniae*, and *Pseudomonas aeruginosa*, and is also effective against certain Gram-positive cocci. It was developed to provide enhanced resistance to aminoglycoside-modifying enzymes compared to its parent compound and was approved for clinical use in China in 2007.
