Drug intelligence / Profile preview
WEX-202 is a topical patch formulation of civamide (also known as zucapsaicin), a potent and selective agonist of the Transient Receptor Potential Vanilloid 1 (TRPV1) receptor. Developed by Winston Pharmaceuticals, the patch was designed to provide localized, sustained relief for chronic pain conditions such as osteoarthritis of the knee and various neuropathic pain syndromes. The mechanism of action involves the initial activation of TRPV1 receptors on nociceptive nerve fibers, which leads to a subsequent prolonged period of desensitization and the depletion of proinflammatory neuropeptides, most notably substance P. This desensitization effectively raises the pain threshold and reduces the transmission of pain signals from the peripheral site of application. WEX-202 was advanced into late-stage clinical development, with an New Drug Application (NDA) filed for the treatment of pain associated with osteoarthritis.
