WG-3D7 is an IgM monoclonal antibody (AβComAb) designed to target specific β-sheet secondary structures present in all Alzheimer's disease (AD) pathologic oligomers. Preclinical studies in mouse models of cerebral amyloid angiopathy (CAA) and AD have demonstrated its efficacy in producing significant cognitive rescue, linked to lowering Aβ oligomers. The drug has been shown to interact with brain vasculature and penetrate the blood-brain barrier without inducing inflammation or microhemorrhages. It reduces soluble aggregated Aβ, as well as vascular and parenchymal Aβ pathology, through an Aβ-sequence independent mechanism. These findings suggest its potential for safe translation to human AD clinical trials with minimal risk of ARIA or autoimmune toxicity.
