WH-4 is an experimental small molecule inhibitor that targets the Signal Transducer and Activator of Transcription 3 (STAT3) signaling pathway. It has been primarily investigated for its potential in treating hepatocellular carcinoma (HCC), particularly as a sensitizing agent to overcome chemoresistance. Research demonstrates that WH-4, especially when combined with sorafenib, additively suppresses the proliferation, colony formation, and migration of liver cancer cells. Its mechanism involves the induction of apoptosis by modulating the Bax/Bcl-2 ratio and the downregulation of ATP-binding cassette (ABC) transporters such as ABCB1 (P-glycoprotein) and ABCG2 (BCRP), which are often responsible for drug resistance. The anti-tumor activity of WH-4 is specifically mediated through the inhibition of the STAT3 pathway, as the overexpression of STAT3 has been shown to abolish its inhibitory effects.
