Overview

WHG626 is an investigational, orally bioavailable small molecule inhibitor of Mucosa-Associated Lymphoid Tissue Lymphoma Translocation Protein 1 (MALT1), developed by Novartis. MALT1 is a paracaspase that plays a critical role in the NF-κB signaling pathway, specifically within the CBM (CARD11-BCL10-MALT1) complex that forms upon B-cell receptor activation. By inhibiting the proteolytic activity of MALT1, WHG626 aims to suppress the constitutive NF-κB signaling that drives the survival and proliferation of various B-cell malignancies. It is currently being evaluated in Phase 1 clinical trials for the treatment of relapsed or refractory Multiple Myeloma, primarily in combination with the BCMA-targeting bispecific antibody WVT078.
