White willow bark (*Salix alba*) is a traditional herbal remedy and dietary supplement primarily used for its analgesic and anti-inflammatory properties. Its primary active constituent is salicin, a glucoside that is metabolized in the body to salicylic acid, the precursor to modern aspirin. Unlike synthetic aspirin, willow bark contains a complex mixture of other salicylates, polyphenols, and flavonoids that may contribute to its therapeutic effects. Its mechanism of action involves the down-regulation of pro-inflammatory mediators, including tumor necrosis factor-alpha (TNF-α) and nuclear factor-kappa B (NF-κB), as well as the reduction of inflammatory cytokines like IL-1beta and IL-6. It is commonly used to manage chronic lower back pain, osteoarthritis, and rheumatoid arthritis.
