Whole glucan particles (WGP) are purified, hollow, porous spheres composed primarily of beta-1,3/1,6-glucan derived from the cell wall of the yeast *Saccharomyces cerevisiae*. WGP acts as a potent immune modulator by serving as a pathogen-associated molecular pattern (PAMP) that is recognized by the Dectin-1 (CLEC7A) receptor on myeloid cells, including macrophages, dendritic cells, and neutrophils. Upon binding, WGP triggers innate immune activation, characterized by the production of pro-inflammatory cytokines and the reprogramming of immunosuppressive myeloid-derived suppressor cells (MDSCs) into an M1-like anti-tumor phenotype. In clinical oncology, WGP is investigated as an adjuvant to enhance the efficacy of monoclonal antibodies and checkpoint inhibitors (such as anti-PD-1 or anti-TIGIT) by priming the innate immune system to coordinate a more robust adaptive T-cell response against tumors.
