Overview

WII-2201 is an orally bioavailable small molecule that acts as a dual inhibitor of phosphoinositide 3-kinase (PI3K) and 3-phosphoinositide-dependent protein kinase 1 (PDK1). Developed by the South Korean biotechnology company Wellmarker Bio, WII-2201 is designed to target the PI3K/AKT/mTOR signaling pathway, which is frequently dysregulated in various cancers. By inhibiting both PI3K and PDK1, the compound aims to achieve more complete suppression of AKT phosphorylation and activity compared to selective PI3K inhibitors, potentially overcoming common resistance mechanisms. The drug is currently being evaluated in clinical trials for patients with advanced solid tumors, with a specific emphasis on colorectal cancer (CRC) patients identified through predictive biomarkers.
