Overview

Wild-type adeno-associated virus-like particles (wtAAVLPs) are non-infectious, DNA-free protein shells derived from Adeno-associated virus serotype 2 (AAV2). These particles are assembled from the major capsid protein VP3 with the assistance of the assembly-activating protein (AAP), forming a stable 25-nm icosahedral structure. Unlike recombinant AAV vectors, wtAAVLPs lack a viral genome and are incapable of gene transfer. They serve as a highly immunogenic scaffold for vaccine development, where foreign epitopes can be inserted into specific capsid positions (such as 453 or 587) to elicit potent B-cell responses. In research contexts, wtAAVLPs are used as negative controls or as the baseline platform for chimeric vaccines targeting pathogens like Human Papillomavirus (HPV). The platform is noted for its stability, which allows for lyophilization and storage without a cold chain.
